چکیده

In macronuclear DNA of the ciliated protozoan Paramecium caudatum, we identified a gene homologous to aubergine/piwi in Drosophila and ZWILLE in Arabidopsis, which have recently been shown to regulate the maintenance of stem cell in animals and plants. The DNA sequence was considered to be actually transcribed because it was also obtained from cDNA of vegetative phase cells by RACE-PCR. The gene named pap (aramecium aubergine/piwi homologue) encodes 781 amino acids and contains three short introns (21bp, 22bp and 23bp) in the ORF. Forty four UAA colons and 8 UAG colons which code for glutamine in Paramecium were found in the ORF. This is the first finding of piwi/ZWILLE homologue in a protozoan. The gene was transcribed actively immediately after induction of conjugation, while the transcription was restricted amount during vegetative phase. The result suggests that the gene is integrated in the developmental process of conjugation.

Response to Morphine in a unicellular animal model (Paramecium caudatum)

Introduction: Response to morphine and role of Nitric Oxide (NO) on expression of morphine response has been studied in vertebrates. But, little evidence is provided in the matter in earlier invertebrates. This investigation for the first time evaluated the effect of NO on expression of morphine potency in Paramecium caudatum. Methods: Animal after isolation from natural media and specific i...

Insights into three whole-genome duplications gleaned from the Paramecium caudatum genome sequence.

Paramecium has long been a model eukaryote. The sequence of the Paramecium tetraurelia genome reveals a history of three successive whole-genome duplications (WGDs), and the sequences of P. biaurelia and P. sexaurelia suggest that these WGDs are shared by all members of the aurelia species complex. Here, we present the genome sequence of P. caudatum, a species closely related to the P. aurelia ...

Acute Toxicity and Cytogenetic Effects of Monocrotophos in Paramecium Caudatum and Oxytricha Fallax

In the present study experiments were conducted to evaluate the toxic effects of monocrotophos on ciliate models Paramecium caudatum and Oxytricha fallax, which can be considered as ideal organisms for investigating water quality fluctuations and risk assessment of pesticides. Cell viability, cell morphology, cell behaviour and macronucleus tests were performed using to different concentrations...

Infection and maintenance of Holospora obtusa, a macronucleus-specific bacterium of the ciliate Paramecium caudatum.

The gram-negative bacterium Holospora obtusa is a macronucleus-specific symbiont of the ciliate Paramecium caudatum, which invades the host cell via a food vacuole, infects its macronucleus and grows exclusively in the nucleus. From infection experiments, we showed that a property of the macronucleus that is necessary for it to be recognized and infected by H. obtusa is commonly provided by P. ...

Coping with Temperature at the Warm Edge – Patterns of Thermal Adaptation in the Microbial Eukaryote Paramecium caudatum

BACKGROUND Ectothermic organisms are thought to be severely affected by global warming since their physiological performance is directly dependent on temperature. Latitudinal and temporal variations in mean temperatures force ectotherms to adapt to these complex environmental conditions. Studies investigating current patterns of thermal adaptation among populations of different latitudes allow ...
